MOLECULAR MECHANISM OF RANKL IN PERIODONTITIS (MEKANISME MOLEKULER RANKL PADA PERIODONTITIS)

Abstract
The mechanism of bone resorption and remodeling is coordinated by the interaction among the receptor activator of nuclear factor-kappa B (RANK), RANK ligand (RANKL), and its decoy soluble receptor osteoprotegerin (OPG). RANKL is a homotrimeric transmembrane protein that is expressed as a membrane bound and a secreted protein which is derived from the membrane form as a result of either proteolitic cleavage or alternative splicing. Many cell types express RANKL, including osteoblasts, periodontal ligament fibroblasts, and endothelial cells. In molecular mechanism of RANKL, RANKL production from fibroblast stimulated by periodontal pathogens, bacterial antigen specific T and B lymphocyte are key players, and there is co-stimulatory molecules. The primary aim of this paper is to explain RANKL-mediated osteoclastogenesis.
